Buchwald配体系列
Buchwald-Hartwig偶联反应是一种在钯催化和碱存在下,胺与卤代芳基、烯烃基和杂环芳基的交叉偶联反应,形成新的C-N键,最终生成胺的N-芳基化产物的反应,是合成芳胺的重要方法。
Buchwald反应的迅速发展,很大程度上归功于新配体的发展和应用。这些富电子的配体L,结构高度可调,为催化剂体系提供多样化的范围、高稳定性和反应活性。这些Buchwald配体是一类联苯类单膦配体,一般为双烷基膦配体(图1)。该类配体通过以下几个方面影响偶联反应的催化活性:1)芳香环2, 6位引入供电性的取代基能够通过防止金属嵌入环化增加催化剂的稳定性,促进零价钯的生成;2)芳基的位阻作用,阻止膦原子被空气氧化,增加了膦配体的稳定性;3)增加苯环上的供电子取代基能够促进催化循环中的还原消除过程;4)环己烷类的大位阻膦配体也能加速还原消除过程,同时这类富电子的基团也能够增加催化循环中氧化加成反应速率。
